On the cover: β-thalassemia is a common monogenic disorder. De-repression of γ-globin to compensate for β-globin deficiency represents a promising genetic therapy for β-thalassemia. Genome-wide association studies have identified transcription factors that drive γ-globin silencing in healthy adults with varying Hb F levels. In this issue, Luo et al. report that a previously unknown SNP, rs7948668, is significantly associated with higher Hb F levels and delays the disease onset (pp. 157–167). The cover image illustrates how the rs7948668 mutation (pink) disrupts the IKAROS binding motif, leading to the dissociation of the PYR Complex (purple). The altered spatial structure of the chromosome (transparent dome) enhances the competition of HBG promoters for locus control regions (red). As a result, γ-globin is activated (gold), ready to synthesize large amounts of fetal hemoglobin.
